Immune modeling is the design of an experimental model to test the immune response associated with a drug, an infection or a metabolite, or a drug. Immuno-oncology can be the main resource to understand the effective pathway of a therapeutic agent, for example an oncolytic agent. This is best suited when looking for a custom solution to a problem that a single or standard test cannot solve.

Navigate and understand the complex immune system with a suitable immune model

An immune model is valuable for developing or testing an immunotherapeutic expected to function by generating an immune response (e.g. against tumours). Or if the drug targets an immune condition (e.g. autoimmunity) or if the drug might generate an adverse immune response. You can also combine this with a mathematical model.

The adaptive immune system is a complex system structured through a cascade of cells, receptors, and cytokines that process, respond, initiate, memorise, and deliver an immune response or tolerance. Therefore, understanding the threshold of ‘Tolerance’ is essential to determine the mechanism of a therapeutic that exerts its effect through an immune response or that is likely to cross paths with the immune system.

In a sterile inflammatory state, as in a drug-induced effect, the immune effectors interact with the body’s tissues and the therapeutic agent to produce a cell-mediated or antibody-mediated immunity.

In addition to the conventional PBMC-based approach, we have developed an in vitro human platform for (co-)cultures of human tissue and immune cells that can be customised to different drug modules to test drug immunomodulatory mechanism and/or immunologic risks. This platform is organic that evolves with our knowledge and experience in immunology.

To study the role of heat shock proteins in immune-mediated cell death and autoimmunity, we designed a diabetes model consisting of transgenic expression of the target protein. We initiated cell death of islet beta-cells in a controlled dose-dependent manner. The study successfully revealed that the (i) initial beta cell death was responsible for initiating immune-mediated diabetes, and (ii) the immune response was increased by the overexpression of heat shock protein. This study was essential to understand the role of heat shock proteins in immunogenicity following tissue damage.
